Optimization of Sliding Indentation Loading to Induce Physiological Collagen Architecture in Growing Tissue Engineered Cartilage

چکیده
Introduction Currently, the load-bearing capacity of tissueengineered cartilage is insufficient. Inducing sufficient collagen content with an arcade-like architecture is a solution. Recently, sliding indentation was proposed for this puropse. However, it was only initially explored and further optimization may yield significantly better results. The aim of this study is to investigate in-silico whether indentation depth of sliding indentation may be optimized to promote the desired collagen orientation in growing agarose-cell constructs.

Sliding contact loading enhances the tensile properties of mesenchymal stem cell-seeded hydrogels.
The primary goal of cartilage tissue engineering is to recapitulate the functional properties and structural features of native articular cartilage. While there has been some success in generating near-native compressive properties, the tensile properties of cell-seeded constructs remain poor, and key features of cartilage, including inhomogeneity and anisotropy, are generally absent in these e...

If a tissue-engineered cartilage transplant is to succeed, it needs to integrate with the host tissue, to endure physiological loading, and to acquire the phenotype of the articular cartilage. Although there are many reported treatments for osteochondral defects of articular cartilage, problems remain with the use of artificial matrices (scaffolds) and the stage of implantation. We constructed ...
متن کاملStick-slip friction and wear of articular joints.
Stick-slip friction was observed in articular cartilage under certain loading and sliding conditions and systematically studied. Using the Surface Forces Apparatus, we show that stick-slip friction can induce permanent morphological changes (a change in the roughness indicative of wear/damage) in cartilage surfaces, even under mild loading and sliding conditions. The different load and speed re...

Purpose: In the present study the effect of BMP-6 was investigated on chondrogenesis of adiposederived stem cells. Materials and Methods: Mesenchymal stem cells derived from subcutaneous adipose tissue were cultured on alginate scaffold to induce chondrogenesis in experimental group, with chondrogenic medium having BMP-6 growth factor for 3 weeks. In control group medium without BMP-6 was appli...
متن کاملPhysiological Collagen Architecture in Engineered Tissue Cartilage through a Combined Approach of Mechanical Stimulus and Anisotropic Fibrous Scaffolds
This work consists on improvement of the functionality of tissue engineered cartilage through a combined approach of mechanical stimulus with anisotropic fibrous scaffolds provided on a bioreactor culture environment, in order to bring the structural organization of engineered cartilage close to the native tissue, reducing the risk of failure of this promising method for osteoarthritis treatment.
